Description:
The series of DNA Size Selection Kits with magnetic beads was designed for precise selection of DNA fragments by size. A total of 11 variants are available, covering a range from 50 bp to over 10 kb. These kits allow for easy and rapid enrichment of samples with DNA fragments of the desired size. The workflow supports both single-sided and double-sided selection, enabling precise size cutoff adjustments. Accurate size selection enhances sequencing efficiency, improves data quality, and reduces costs.
Magnetic bead technology uses paramagnetic particles, also known as SPRI (Solid Phase Reversible Immobilization) beads, which are used for reversible and selective DNA binding. DNA fragments can be selectively chosen and purified by altering the properties of the magnetic or SPRI beads. Magnetic beads easily separate the DNA bound to the beads from contaminants and unwanted components in the samples. After DNA size selection, the samples are free of contaminants such as buffer components, enzymes, proteins, salts, dNTPs, primers, and adapters. Our proprietary magnetic bead reagents improve yield, selectivity, and reproducibility.
Specific DNA fragments of a certain length can be easily purified using magnetic separation with different bead components, eliminating the time-consuming and complex methods such as gel extraction or column purification. The magnetic bead method is popular for routine DNA size selection, including library size selection.
Procedure:
The double-clean-up DNA selection procedure is illustrated in the image below. The first bead-binding step, referred to as right-side clean-up, removes large DNA fragments. These large fragments bind to the beads and are then discarded. The desired DNA fragments in the supernatant are transferred to a new well, and new beads are added for the second bead-binding, called left-side clean-up. The double-size-selected DNA fragments are eluted after ethanol washing.
