Description:
Proteinase K IVD (lyophilisate) is an enzyme for digesting proteins in biological samples with an activity of ≥30 U/mg, intended for in vitro diagnostic use. Proteinase K (EC 3.4.21.64) is a broad-spectrum serine protease. The predominant site of cleavage is the peptide bond adjacent to the carboxyl group of aliphatic and aromatic amino acids with blocked alpha amino groups. It is a recombinant enzyme cloned from the fungus Engyodontium album and produced in Pichia pastoris. It is inactivated by diisopropyl fluorophosphate (DFP) or phenylmethanesulfonyl fluoride (PMSF). It is supplied as a lyophilisate, which is dissolved in sterile water or in the recommended storage buffer before use.

Features:

  • Recombinant proteinase K (EC 3.4.21.64), a broad-spectrum serine protease
  • Activity ≥30 U/mg
  • Form: lyophilisate
  • Recombinant enzyme from the fungus Engyodontium album produced in Pichia pastoris
  • Inactivation by DFP or PMSF
  • Dissolved in sterile water or the recommended storage buffer (20 mM Tris, pH 7.5, 1 mM CaCl₂, 0.02% sodium azide, 50% glycerol), which are not included
  • For long-term storage of the solution, dissolving the lyophilisate in the buffer above is recommended
  • At concentrations below 1 mg/ml the enzyme undergoes natural autoproteolysis
  • Storage of the lyophilisate and the prepared solution at 2–8 °C, transport at 15–25 °C
  • For in vitro diagnostic use (IVD)

Applications:

  • Proteolytic degradation of biological material
  • Inactivation of endogenous DNases and RNases during nucleic acid purification

Kit contents:

  • Proteinase K, recombinant, lyophilisate (storage at 2–8 °C)
