PcrA Helicase

ATP-dependent 3′ to 5′ helicase

expressed in E. coli

Description:
PcrA is an ATP-dependent 3′ to 5′ helicase responsible for unwinding double-stranded DNA (dsDNA). PcrA binds to dsDNA and separates it into two single-stranded DNA (ssDNA) molecules.

The enzyme is supplied at 100 μg, 0.5 mg/ml.

10X Reaction Buffer (500 mM Potassium acetate, 200 mM Tris-acetate, 100 mM Magnesium acetate, 1 mg/ml BSA, pH 7.9)

Applications:
Helicase-dependent DNA isothermal amplification.

Experiment Result:

Helicase-dependent DNA isothermal amplification reaction (65 °C, 1 hour) using PcrA helicase for cloning 667 bp and 871 bp DNA fragments. Template of the reaction is 5 ng DNA.
