Description:

Blunt/TA Ligation Master Mix is a ready-to-use 2X master mix designed for the efficient joining (ligation) of challenging DNA fragments – both blunt-ended and those with 1-base overhangs. It contains T4 DNA ligase and optimized components that significantly enhance the efficiency and speed of ligation, even with demanding samples.

Blunt/TA Ligation Master Mix can ligate the following types of DNA fragments:


Ligace DNA

The image shows the result of ligating DNA fragments with single-base overhangs using the Blunt/TA Ligation Master Mix under different reaction times at 20 °C (left) or different temperatures for 15 minutes (right). The number of transformants obtained from triplicate plating demonstrates that the Master Mix works very well at room temperature (20 °C–25 °C) in just 15 minutes.

The graph shows a comparison of T4 DNA ligase (high concentration), Rapid Ligation Master Mix (Cat.# 40102), and Blunt/TA Ligation Master Mix (Cat.# 40101) for ligation of nicked, sticky-end, 3′ single-base overhang, and 5′ single-base overhang substrates. All ligations were incubated for 15 minutes, except for the 5′ single-base overhang, which was incubated for 1 hour. Blunt/TA Ligation Master Mix achieves the highest efficiency not only with nicks and sticky ends (up to 90%) but also significantly outperforms standard T4 ligase with fragments containing 3′ and 5′ single-base overhangs.
