Quantification of oligo < 50 nt

For use with Qubit™ fluorometer

Description: 

BioDynami DNA/RNA quantification kits are designed for accurate quantification of DNA and RNA samples across a wide concentration range using the Qubit™ fluorometer.

The Short Oligo Quantification Kit is developed for quantification of oligos shorter than 50 nt and includes dye, dilution buffer, and two standards. The assay is accurate for oligo concentrations ranging from 50 pg/µL to 200 ng/µL based on comparison of the data with standards, as shown in the graph below.

Protocol:

The test procedure is simple – just dilute the dye with the dilution buffer, add the oligo sample (volume 1–20 μL), and then measure the oligo concentration using the Qubit® fluorometer.

  • Uses the Qubit™ ssDNA assay setting
  • Linear range: 1–200 ng oligo

Quantification:

Long oligos (>50 nt) and ssDNA can be quantified using ssDNA Quantification kits. However, quantifying the concentration of oligos shorter than 50 nt is challenging due to data inaccuracy. Therefore, the Short Oligo Kit was developed, which is the most accurate for quantifying short oligonucleotides, especially for very short sequences (11 nt), where other kits fail. A comparison of the kits can be seen in the graph below.

Effect of Contaminants:

The tests are well-tolerated by common contaminants such as salt, solvents and detergents as shown in the table below.
