Description:

The Library Size Selection Kit series from the U.S.-based company BioDynami is designed for NGS library preparation for both short- and long-read sequencing. This kit is most suitable for selecting library sizes for long-read sequencing with DNA fragments larger than 10 kb.

The workflow supports both double-sided size selection for short reads and single-sided selection for long reads. Thorough and high-quality library preparation is essential for achieving accurate and reliable sequencing results. Precise fragment size selection increases sequencing efficiency, improves data quality, and reduces costs.

The magnetic bead technology utilizes paramagnetic particles, also known as SPRI beads (Solid Phase Reversible Immobilization), which enable reversible and selective DNA binding. DNA fragments can be selectively captured and purified by modifying the properties of these beads. The magnetic beads efficiently separate DNA from contaminants and unwanted components such as buffer components, enzymes, proteins, salts, dNTPs, primers, and adapters. The kit's patented reagents enhance yield, selectivity, and reproducibility, ensuring optimal performance.

Procedure:
Library size selection for long-read sequencing requires only a single clean-up step, as shown in the image below. During this process, large library fragments bind to the beads, while smaller fragments remain in the supernatant and are discarded. After the rinsing step, the selected larger fragments are eluted in water or an appropriate buffer.
