Magnetic beads for tRNA purification from the American company BioDynami combine BioDynami's patented chemistry with the unique properties of SPRI beads (Solid Phase Reversible Immobilization) and were developed to address the challenges of tRNA and short DNA/RNA fragment purification.
They are optimized for efficient tRNA cleaning from impurities and unwanted components such as dNTPs, detergents, salts, proteins, and other contaminants. The reagents can be used for applications with oligonucleotides (>70 nt).
Procedure:
The tRNA purification process using magnetic beads is illustrated below with a sample containing long DNA/RNA fragments (>180 nt). The purification begins with the binding of large RNA/DNA molecules (green) to the magnetic beads (brown) in a solution containing tRNA (blue) and impurities (yellow). Using a magnet, the beads with bound RNA/DNA are separated from the solution, removing impurities. tRNA then binds to the beads, followed by another magnet separation to remove undesired material. After washing, the remaining impurities are removed, and in the final elution step, RNA/DNA is released from the magnetic beads into a clean solution.

Efficiency:
The graph shows that BioDynami magnetic beads effectively purify tRNA and 70 nt oligo, with high recovery rates for both nucleic acid types. The recovery rate difference between tRNA and 70 nt oligo is relatively small, suggesting that the beads have good affinity for both types of molecules.
Gel electrophoresis shows that the product enables the acquisition of cleaner RNA samples, minimizing contamination from ribosomal RNA and large fragments. This is crucial for subsequent applications where RNA quality and purity are essential.
