PRINCIPLE
The peqGOLD XChange Plasmid Midi kits
combine a modified method of alkaline
lysis with the DNA binding properties of
XChange Anion exchange gels.
An optimized packing process leads to
a porous column matrix where bacterial
lysates or Wash and Elution buffers can
rapidly and consistently pass through by
gravitation. The column matrix contains
positively charged active groups which
bind the negatively charged DNA while
enzyme inhibitors and contaminants
are easily washed through. After the
washing step the pure plasmid DNA is
eluted and then alcohol precipitated.
Although flow rates are rapid and protocol
times are short, the isolated DNA
is highly pure, provides excellent chromatographic
definition and thus is ready
to use for a wide range of downstream
applications.
