Magnesium is the cofactor for the polymerase. This patentpending technology relates to thermally reversible chelators of magnesium, thus a hot-start-cold-stop PCR is achieved.
At lower temperature below 37oC, magnesium is chelated and not able to bind the polymerase. At elevated temperature above 65 or 72oC, magnesium is released to bind the polymerase. The chelators will compete for the magnesium back from the polymerase when the temperature lowered below 37oC.
This mechanism provides a thermally reversible Hot-start PCR system without any modification on polymerase itself. This hot-start PCR system requires no heat activation step.
